General- Updated 4/2/19 CG
BR Project
- Factor VIII project temporarily on hold. CG completed a 20 page report per BR's request. Biorad reviewing report to determine next steps.
- AEX Tech Transfer run did not go well. Still waiting to hear back from Biorad
TG Project
- Tubing and transfer caps prepped and autoclaved
- Will need to make buffers and media this week
- Mariusz to drop off a large (XK26 or XK50) chromatography column this week to be using downstream.
- Currently scaling up for 10L bioreactor. Need 1500mL of culture at around 2E6vc/mL to seed bioreactor at 0.4E6vc/mL.
- Inoculation of 10L Bioreactor scheduled for Tuesday 4/9.
- Will start with 7L working volume
- Feed 1 on 4/12 (Day 3) will be 1L of CD Efficient Feed
- Feed 2 on 4/15 (Day 6) will be 2L of CD Efficient Feed
- Will take several 1mL samples daily
- 1mL for ELISA (freeze immediately)
- 1mL for Western Blot (freeze immediately)
- 1mL for offline cell count
- 1mL for offline pH (pH strips), glucose, and lactate measurements
AM Project
- More AM projects in the pipeline, targeting 1-3 April projects.
- Repeat of STEM with some modifications
- Many repeats (16 samples) of PRO with modifications
- New project with similar protocol and still in hUCMSC's (BIOG)
LYO (TM)
- Still trying to test vacuum leak
- Pending functional vacuum hold, readiness activities to re-inititiate (cleaning/microbial testing 1'st, followed by mock run with mannitol buffer)
