ABSTRACT:

Research in my laboratory is broadly focused on understanding the structure and function of macromolecular machines using cryo-electron microscopy (cryo-EM). There are two specific avenues of investigation: (1) mechanisms of RNA-guided CRISPR-Cas surveillance CRISPR-Cas surveillance complexes and (2) methods to solve structures of human complexes in a highly parallel manner. I will present recent insights into the assembly of CRISPR-Cas effector complexes and implications for target processing. I will also discuss the progress that we are making towards developing algorithms for reconstructing and building models of complexes from the lysate of a single cell.
